Studies on effects of Histamine on Renal Functions

Abstract
The direct effect of histamine on renal function in anesthesized dogs were studie.
Mongoral dogs weighing 10¡­12kg were anesthetized with 30mg/kg of pentobarbital sodium.
The right femoral artery and vein were cannulated with polyetylene tubing to facilitate withdrowal of blood samples and infusion of fluids, respectively.
Both ureters were cannulated with polyethylene tubing and urine from the right and left kidneys was collected separately. The right kidney served as a control. Into the left renal artery a 22 gauge needle was introduced for histamine administration.
The animals were hydrated, intravenously, by a sustaining infusion of 0.9% saline, containing 0.2% of inulin so that the rate of glomerular filtratin could be estimated, at the rate of 0.3§¢/kg/min. Para-aminohippuric acid (PAH) (0.4mg/kg/min) was added to the latter of infusion so that the rate of renal plasma flow could be estimated.
The urine were colleted from the period of 90minute after initiation of infusion of saline.
In this studies the dogs were divided into six groups, consistion of five dogs each, according to dose and drugs used.
Group 1. (control) : 0.9 per cent NaCl solution were infused into left renal artery,
Group 2. (histamine 0.1 ug group) ; histamine 0.1 ug/kg/min. were infused.
Group 3. (histamine 0.5 ug group) ; histamine 0.5 ug/kg/min. were infused.
Group 4. (histamine 1.0 ug group) ; histamine 1.0 ug/kg/min. were infused.
Group 5. (histandne 1.5 ug group) : histamine 1.5 ug/kg/min. were infused.
Group 6. (histamine+Furosemide group) histamine 0.5 ug/kg/min with furosemide 10 ug/kg/min. were infused.
Histamine in normal saline solution were infused into the left renal artery and comparison of urinary output renal plasma flow (RPF), glomerular filtration rate (GFR), urinary sodium and potassium of both kidney were made.
The results obtain in this study were as follows:
1) In histamine 0.1 ug group, urinary output, renal plasma flow (RPF), glomerular filtration rate (GFR) urinary sodium and potassium were increased slightly, but in the groups of histamine 0.5 ug and 1,0 ug increased markedly on the histamine injected side.
2) In histamine 1.5 ug group, urinary output, RPF, urinary sodium and potassium of histamine injected side were increased markedly, but these of the other side were decreased only slightly.
3) In histamine+Furosemide group urinary output, RPF, GFR, urinary sodium and potassium of the injected kidney were increased and those diuretic action of furosemide and furosemide shows additive.
